Abstract
The humeral response of 32-day-old specific-pathogen-free Fisher 344 strain inbred albino female rats to a 5,000 sporulated oocyst inoculum of Eimeria nieschulzi at postinoculation day 0 (PI+0) was followed for 77 days, as were syngeneic uninfected control rats of the same age. Some of these infected rats were challenged with 50,000 sporulated oocysts at PI+15 and followed through PI+77; some were challenged with 50,000 sporulated oocysts at PI+15 and PI+60 and followed through PI+77; and some were challenged with 50,000 sporulated oocysts at PI+60 and followed through PI+77. The Pfizer agarose system of analysis was used for the serum and the Biogel A1.5M column fractionation system of analysis was used for the unpurified coprantibody preparation, with subsequent Pfizer agarose system additional immunoelectrophoretic analysis. No observable significant differences were noted between infected (challenged or not) rats and uninfected syngeneic control rats during this 77-day period. There is a very strong implication that future humeral response experiments should be conducted with rats not less than 100 days of age at the onset of experimentation. Unsuccessful attempts were made to transfer passive immunity to 32-day-old rats beginning PI-2 by repeated intraperitoneal injections of freshly thawed immune serum or by repeated inoculations per os of freshly resuspended lyophilized immune unpurified coprantibody preparation beginning PI-1. No immunologically significant observable differences were noted during this 17-day period in oocyst production, sporulation, patency, serum components, or unpurified coprantibody preparation fractionation. Present results add another link to the immunological chain of events that lead to immunity to Eimeria nieschulzi and further support the view that this protective immunity is primarily a cell-mediated response.
